Small-conductance Ca activated K channels modulate action potential-induced Ca transients in hippocampal neurons. J Neurophysiol 109: 1514–1524, 2013. First published December 19, 2012; doi:10.1152/jn.00346.2012.—In hippocampal pyramidal neurons, voltage-gated Ca channels open in response to action potentials. This results in elevations in the intracellular concentration of Ca that are maximal in the proximal apical dendrites and decrease rapidly with distance from the soma. The control of these action potential-evoked Ca elevations is critical for the regulation of hippocampal neuronal activity. As part of Ca signaling microdomains, small-conductance Ca -activated K (SK) channels have been shown to modulate the amplitude and duration of intracellular Ca signals by feedback regulation of synaptically activated Ca sources in small distal dendrites and dendritic spines, thus affecting synaptic plasticity in the hippocampus. In this study, we investigated the effect of the activation of SK channels on Ca transients specifically induced by action potentials in the proximal processes of hippocampal pyramidal neurons. Our results, obtained by using selective SK channel blockers and enhancers, show that SK channels act in a feedback loop, in which their activation by Ca entering mainly through L-type voltage-gated Ca channels leads to a reduction in the subsequent dendritic influx of Ca . This underscores a new role of SK channels in the proximal apical dendrite of hippocampal pyramidal neurons.

متن کاملFunctional roles of SK2 channels in area CA1 of the hippocampus
Ca+-activated small-conductance K+ (SK) channels play a fundamentally important role in all excitable cells. They are potassium selective, voltage-insensitive and are activated by increases in the levels of intracellular Ca+ such as occur during an action potential (Zhang et al., 1995; Kohler et al., 1996; Sah et al., 1991; Lorenzen et al., 1992). As the action potential decays, the membrane po...
متن کاملActivation by zonisamide, a newer antiepileptic drug, of large-conductance calcium-activated potassium channel in differentiated hippocampal neuron-derived H19-7 cells.
Zonisamide (ZNS; 3-sulfamoylmethyl-1,2-benzisoxazole), as one of the newer antiepileptic drugs, has been demonstrated its broad-spectrum clinical efficacy on various neuropsychiatric disorders. However, little is known regarding the mechanism of ZNS actions on ion currents in neurons. We thus investigated its effect on ion currents in differentiated hippocampal 19-7 cells. In whole-cell configu...
متن کاملParadoxical role of large-conductance calcium-activated K+ (BK) channels in controlling action potential-driven Ca2+ entry in anterior pituitary cells.
Activation of high-conductance Ca(2+)-activated K(+) (BK) channels normally limits action potential duration and the associated voltage-gated Ca(2+) entry by facilitating membrane repolarization. Here we report that BK channel activation in rat pituitary somatotrophs prolongs membrane depolarization, leading to the generation of plateau-bursting activity and facilitated Ca(2+) entry. Such a par...
